Muscadine grapes have shallow roots that feed within the top 12 inches of the soil. I like to fertilize mine now and again in late June. A good soil test is helpful to determine the fertilization needs. Magnesium is an important nutrient for Muscadines. Ag. lime is a good source for that but be careful not to raise the ph much above 6 (5.8 to 6.5 preferred) .

Meadowlark is sure right about getting a soil test before adding any fertilizer to your plants. That is what makes a difference between someone that knows what they are doing and someone that don't. I think getting a soil test is about the most important thing a person can to for their gardens at least every 5 years. I get mine with the best recommendations from the soil pros at Texas A&M for $27.
